Selection of the mutants was done by amplifying the region containing sgRNAs 8, 1 and 7 using flanking primers Fw2969 and Rv4230 (Fig. 1). A 1262-bp wild-type allele was amplified in Moneymaker (MM) and transformant TV161196 (non-mutant). Smaller fragments than the wild-type allele indicate deletions between sgRNAs.
