Quantitative RT-PCR analysis of gene expressions in dry and imbibed seeds of the BnPHT1;4 overexpression transgenic Brassica napus. Gene expression of BnPHT1;4, GA-related genes (BnGA1, BnGA2, BnGA3 and BnKAO1), ABA biosynthesis-related genes (BnNCED5 and BnNCED9) and ABA signaling positive regulator gene (BnABI4) was determined in germinating seeds during the course of the imbibition process. Two-month stored seeds were used for seed germination, and total RNA was isolated from the dry and imbibed (germinating) seeds for RT-PCR analysis, using BnACT2 gene as standard control. The data are presented as means ± SD (n = 3), and significance of difference was analyzed by Duncan’s test.
