Schematic of peptidomic analysis through the one-pot derivatization of cysteine to pseudo-lysine. Plant extract was fractionated by HPLC and analysed by MALDI-MS. Fractions with peaks between 2000 to 4000 Da were pooled and lyophilized. The dried sample was mixed with dithiothreitol (DTT) and 2-bromoethylamine and incubated at pH 8.6 at 55 °C for 1 h. The reaction mixture was subjected to Orbitrap MS and the fragmentations were matched with the transcriptomic data to confirm the sequences
