Effect of HRW, SNP, cPTIO, L-NAME and NaN3 on cell cycle phase distribution during adventitious rooting. The primary root system was removed from hypocotyls of 5-day-old germinated cucumber. Explants of cucumber were incubated with 50 % HRW, 50 μM SNP, 200 μM cPTIO, 30 μM L-NAME or 10 μM NaN3 as indicated for 6 h. Cucumber hypocotyls were fixed by paraformaldehyde and nuclei were extracted by chopping in Galbraith buffer. The nuclei were resulted with DAPI, and then visualized by fluorescent microscopy; the same samples were processed for multiparametric cell cycle high-content analysis (HCA) (a). Bar = 100 μm.
