EpiBL treatment of wheat embryos. Embryos were isolated out of the kernel at sizes ranging from 100 to 280 μm in diameter, most of them having a diameter of 120 to 180 μm. They were then grown in vitro on media supplemented with 0 to 10 μM epiBL. The phenotypes of the in vitro grown embryos were observed after 7 to 8 days of culture and grouped into classes. Photographs of representative phenotypes after 7/8 days of culture were added directly to the figure (a-f, 1photographs not shown). Reported numbers in (g) are percentages of defined phenotypes per growing embryos. In at least 4 independent experiments, a total of 62 growing embryos were observed in control conditions, 41 for 0.5 μM epiBL, 33 growing embryos for 1 μM epiBL, 35 in the case of 2 μM epiBL, 34 for 5 μM epiBL and 29 in the case of 10 μM epiBL. apex: differentiation of a leaf primordium or/and a coleoptilar ring (both in most cases abnormal) in addition to a shoot meristem; cr: coleotilar ring; RG: Radial Growth (BS: Ball-Shaped or PS: Pear-Shaped); sc or scut: scutellum; sm: shoot meristem; sp: suspensor; wt: embryo having a wild type phenotype. The percentage of indifferentiated, abnormal or early arrested embryos for a given concentration and a given embryo size range is obtained by subtracting the sum of all indicated percentages to 100 %
