From: EsDREB2B, a novel truncated DREB2-type transcription factor in the desert legume Eremosparton songoricum, enhances tolerance to multiple abiotic stresses in yeast and transgenic tobacco

Figure 5

Growth of S. cerevisiae yeast cells transformed with the empty vector (1) PYES2 and with the (2) PYES2-EsDREB2B under different stress conditions. To test the osmotic and salt tolerances, the same quantity of yeast culture sample was re-suspended in 30% PEG6000 and 5 M NaCl, respectively, at 30°C for 36 h. To test the cold and heat tolerances, the yeast cells were re-suspended in SC-ura medium and placed either in a refrigerator at -20°C alcohol bath or in a water bath at 53°C, respectively, for 24 h. To test oxidative tolerance, yeast cells were incubated in 20 mM H2O2 at 30°C for 8 h. For non-stress, control (NS), an equivalent number of yeast cells was re-suspended in 200 μl of sterile water and incubated at 30°C for 36 h. Serial dilutions of 1:10 transformed yeast cells were grew on SC-Ura medium for 3 d. A non-stress (NS), B osmotic stress, C salt stress, D cold stress, E heat stress and F oxidative stress.
