Laser microdissection of epidermal and cortical cells from rice roots. (A) Typical transverse section from the rice root. Two independent experiments were carried out for isolation of epidermal and cortical rice root cells by LMD. (F) RT-PCR analysis to detect OsCPK18 transcripts in laser microdissected cells from rice roots. Cells were harvested from G. intraradices-inoculated (+Gi) and mock-inoculated (-Gi) roots. Total RNA samples were obtained from pooled microdissected cells. Expression analysis was carried out using the one-step procedure for RT and PCR amplification. OsCPK18 transcripts were detected only in cortical cells. Bars, 100 μm (A), 50 μm (B to E).
