Two independent experiments were carried out for isolation of epidermal and cortical rice root cells by LMD. (F) RT-PCR analysis to detect OsCPK18 transcripts in laser microdissected cells from rice roots. Cells were harvested from G. intraradices-inoculated (+Gi) and mock-inoculated (-Gi) roots. Total RNA samples were obtained from pooled microdissected cells. Expression analysis was carried out using the one-step procedure for RT and PCR amplification. OsCPK18 transcripts were detected only in cortical cells. Bars, 100 μm (A), 50 μm (B to E).
